Hold the position for a count of five to ten and release down to the floor.



2. Squats – Squats work the butt, the hamstring muscles and the quadriceps. If you aren’t sure of proper form, you can use a chair. Stand with feet shoulder width apart and feet firmly planted. Push your butt back as if you were preparing to sit in a chair. Keep your abs tight and your upper body straight. Once you reach chair level stop and hold the position for a count of two to five and release. At the lowest point, place all of your weight on your heels for balance and maximum toning.



3. Reverse Lunges – Lunges work the quadriceps muscles. They can be hard for people with knee problems. A reverse lunge still tones the right muscle groups but with less pressure on the knee. Stand with feet together and arms at your sides. Take one leg and move it backwards until you are in lunge position: front leg bent at a 90 degree angle and back leg extended until you are on the ball of your foot. From this position lower yourself down until the back knee almost touches the floor. Hold for a count of two and return to starting position.



4. Pushups – This is a classic toning exercise that works all areas of the arms plus the chest muscles. If you aren’t comfortable or strong enough to perform a pushup on your toes, lower your body to your knees. Be sure your arms are tucked into the body and your back straight as you lower and lift your body.



5. Crunches – Abdominal muscles can be worked every day to build strength and muscle tone. Lying on the floor in sit up position, lace your fingers behind your head. Squeezing your abdominal muscles, lift your upper body until your lower back is about to come off the floor. Hold for two to five counts and return to starting position.

Researchers Reprogram Brain Cells To Become Heart Cells



For the past decade, researchers have tried to reprogram the identity of all kinds of cell types. Heart cells are one of the most sought-after cells in regenerative medicine because researchers anticipate that they may help to repair injured hearts by replacing lost tissue. Now, researchers at the Perelman School of Medicine at the University of Pennsylvania are the first to demonstrate the direct conversion of a non-heart cell type into a heart cell by RNA transfer. Working on the idea that the signature of a cell is defined by molecules called messenger RNAs (mRNAs), which contain the chemical blueprint for how to make a protein, the investigators changed two different cell types, an astrocyte (a star-shaped brain cell) and a fibroblast (a skin cell), into a heart cell, using mRNAs.

James Eberwine, PhD, the Elmer Holmes Bobst Professor of Pharmacology, Tae Kyung Kim, PhD, post-doctoral fellow, and colleagues report their findings online in the Proceedings of the National Academy of Sciences. This approach offers the possibility for cell-based therapy for cardiovascular diseases.

"What's new about this approach for heart-cell generation is that we directly converted one cell type to another using RNA, without an intermediate step," explains Eberwine. The scientists put an excess of heart cell mRNAs into either astrocytes or fibroblasts using lipid-mediated transfection, and the host cell does the rest. These RNA populations (through translation or by modulation of the expression of other RNAs) direct DNA in the host nucleus to change the cell's RNA populations to that of the destination cell type (heart cell, or tCardiomyocyte), which in turn changes the phenotype of the host cell into the destination cell.

The method the group used, called Transcriptome Induced Phenotype Remodeling, or TIPeR, is distinct from the induced pluripotent stem cell (iPS) approach used by many labs in that host cells do not have to be dedifferentiated to a pluripotent state and then redifferentiated with growth factors to the destination cell type. TIPeR is more similar to prior nuclear transfer work in which the nucleus of one cell is transferred into another cell where upon the transferred nucleus then directs the cell to change its phenotype based upon the RNAs that are made. The tCardiomyocyte work follows directly from earlier work from the Eberwine lab, where neurons were converted into tAstrocytes using the TIPeR process.

The team first extracted mRNA from a heart cell, then put it into host cells. Because there are now so many more heart-cell mRNAs versus astrocyte or fibroblast mRNAs, they take over the indigenous RNA population. The heart-cell mRNAs are translated into heart-cell proteins in the cell cytoplasm. These heart-cell proteins then influence gene expression in the host nucleus so that heart-cell genes are turned on and heart-cell-enriched proteins are made.

To track the change from an astrocyte to heart cell, the team looked at the new cells' RNA profile using single cell microarray analysis; cell shape; and immunological and electrical properties. While TIPeR-generated tCardiomyocytes are of significant use in fundamental science it is easy to envision their potential use to screen for heart cell therapeutics, say the study authors. What's more, creation of tCardiomyoctes from patients would permit personalized screening for efficacy of drug treatments; screening of new drugs; and potentially as a cellular therapeutic.

These studies were enabled through the collaboration of a number of investigators spanning multiple disciplines including Vickas Patel, MD and Nataliya Peternko from the Division of Cardiovascular Medicine, Miler Lee, PhD and Junhyong Kim, PhD from the Department of Biology and Jai-Yoon Sul, PhD and Jae Hee Lee, PhD also from the Department of Pharmacology, all from Penn. Research Targets Malignant Glioma Stem Cells



Cleveland Clinic researchers have identified a cellular pathway that cancer stem cells use to promote tumor growth in malignant glioma, an aggressive brain tumor. The research - published in Cell - also found that existing medications block this cancer-promoting pathway and delay glioma growth in animal models, suggesting a new treatment option for these often fatal brain tumors.

Malignant gliomas account for more than half of the 35,000-plus primary malignant brain tumors diagnosed each year in the United States. Unfortunately, the outlook for patients with malignant gliomas is poor. For patients with the most severe, aggressive form of malignant glioma (grade IV glioma or glioblastoma multiforme), median survival is 9 to 15 months with the best available therapies. These treatments include surgery followed by radiation therapy with the chemotherapy temozolomide followed by additional temozolomide treatment.

Although differences in tumors between people were known to exist, researchers have only recently begun to understand the importance of differences between cancer cells within the same patient. Groups of cells within a glioma which promote brain tumor formation in animal models - called cancer stem cells - have been identified. These cancer stem cells are often resistant to radiation and chemotherapy, making them an important target for developing new and effective disease treatments.

In a recently published manuscript, a team of Cleveland Clinic researchers - led by Jeremy Rich, M.D., Chairman, and Anita Hjelmeland, Ph.D., of the Department of Stem Cell Biology and Regenerative Medicine of the Lerner Research Institute of Cleveland Clinic - define a novel molecular pathway that cancer stem cells use to promote tumor growth. Cancer stem cells produce elevated nitric oxide, a molecule whose role in cancer is not well defined but which has been linked to therapeutic resistance, evasion of cell death, and enhanced proliferation. Nitric oxide is produced in cancer stem cells through increased levels of the enzyme nitric oxide synthase 2 (NOS2); decreasing the level or activity of this enzyme reduces cancer stem cell growth. When drugs inhibiting the NOS2 enzyme were given to mice with gliomas, the growth of the tumors was delayed. Using the National Institutes of Health supported database of glioma specimen data, they also found that increased levels of this enzyme are associated with decreased survival of glioma patients. Targeting this pathway could therefore provide benefits for glioma patients.

Current therapies for cancer have many side effects, because they rely on the increased sensitivity of the cancer cells to drugs which are toxic to normal cells. It is therefore important to consider the potential side effects of any new proposed cancer treatment. The current study did compare the levels of the enzyme NOS2 and the effects of its inhibitors in cancer stem cells and normal neural stem cells. Normal neural stem cells have some similar properties as cancer stem cells from gliomas but cannot form tumors. Only cancer stem cells were found to depend on the enzyme for their growth. Drugs inhibiting the NOS2 enzyme were evaluated in patients for the treatment of other diseases and had minimal toxicity. The authors suggest that these drugs may therefore be useful for enhancing the effects of current therapies without adding a lot of side effects. NOS2 inhibitors may thereby pose a potential therapeutic breakthrough for this lethal disease.

I can't say enough about meditation and prayer. Study after study have proven a reduction in every skin and body malody even cancer symptoms from people that pray and meditate daily. Take 20 minutes every day and do this you will also live longer.

Professor Macchiarini, who works at the of Karolinska University Hospital and Karolinska Institutet, led an international team including professor Alexander Seifalian from the UCL (University College London, UK) and Harvard Bioscience (Boston, USA).

Professor Alexander designed and built the nanocomposite tracheal scaffold, and Harvard Bioscience produced a specifically designed bioreactor used to seed the scaffold with the patient

These are specially trained people who are willing to come to your house to show you how to correctly exercise and they do not cost an arm and a leg.

If you have attempted this route before but have had a negative experience do not despair. It may well be because you just did not hit it off with your personal trainer. I have been using a personal trainer for the last five months and only plumped for him after trying out four others first.

My personal trainer is very good for me. He keeps me on my toes, is very good at making me continue to work hard when all I want to do is quit. He is also very fair and seems to understand how far to push me. Together we have worked out and agreed a program which includes various goals for the future. He seems to also understand about the types of food which I should and should not be eating. I am happily still able to eat a lot of foods which I like and he has also introduced me to a number of other ideas which are healthy but also very tasty.

Hiring a personal trainer was the best thing I have ever done with regards to losing weight and I would recommend it to other people. I have now lost the beer belly and still continue working hard, it is now more as a fitness thing rather than to lose even more weight.

The trainer has even stated that I am likely to actually gain weight in the future as certain muscles grow. I will not be fat though, I will be toned and a little but muscular. I look forward to the future and can also now for the first time in a number of years enjoy shopping for clothes.

Embryonic Stem Cells Used In New Model For Studying Germ Cell Tumors In Testes



A team of researchers from Spain and Switzerland have developed a new model for studying the development of testicular germ cell tumors by transplanting embryonic stem cells into the seminiferous tubules in mouse models, resulting in the development of testicular germ cell tumors (TGCT) that mimic the early stages of TGCT development. The study, published in Cell Transplantation (20:5), is now freely available on-line here.

"Over the last fifteen years, cell transplantation into seminiferous tubules has become a valuable tool for studying germinal cell biology," said the study's corresponding author Dr. Juan Arechaga of the University of Basque Country, Spain. "The blood-testes permeability barrier establishes a sealed compartment that protects against influences such as immunological rejection. Thus, our lab has developed a tumor assay to study the cancer invasion process in testicular germ cell tumors using embryonic stem cells injected into the seminiferous tubules."

According to Dr. Arechaga, the tumors generated by the transplantation of early embryos possess gene expressions and differentiation patterns similar to those in testicular germ cell tumors.

The researchers point out that testicular germ cell tumors are uncommon in non-human animals, so there have been no previously suitable animal models for study. However these tumors appear to be genetically regulated and the specific genes related to the development of this kind of tumor have been identified.

"Embryonic stem cell transplantation into mouse seminiferous tubules represents a model with very valuable potential applications because it mimics the pre-invasive state of TGCTs," said Dr. Arechaga. "Donor cells can be transfected with different transgenes before transplantation to evaluate certain genes during the invasive process."

The authors also note that the approach could also be useful in screening novel therapeutic drugs, including inhibitors of angiogenesis and metastasis, to potentially treat TGCTs.

"This study demonstrates the development of an animal model of testicular germ cell tumor formation" said Dr. David Eve, associate editor of Cell Transplantation and Instructor at the Center of Excellence for Aging and Brain Repair, University of South Florida. "Study of this model will further our understanding of how testicular cancers arise and potential ways to treat it."

Neural Stem Progenitor Cell Transplantation's Potential To Aid Spinal Cord Injury Tested



A study published in the current issue of Cell Transplantation (20:5) investigating optimal routes for transplanting neural stem/progenitor cells (NS/PCs) in animal models of spinal cord injury (SCI) has demonstrated that intralesional (IL) injection conferred benefits over intravenous injection (IV) and intrathecal (IT) injection. The study, by a team of Keio University (Japan) researchers, is now freely available on-line here.

"Spinal cord injury usually results in severe, paralytic damage because the adult central nervous system has little potential for regeneration after injury," said corresponding author Dr. Masaya Nakamura of the Keio University School of Medicine's Department of Orthopedic Surgery. "With cell transplantation of neural stem/progenitor cells being a promising strategy, our study sought to determine the best method of application for optimal cell survival and subject safety."

For their study, the researchers used a new reporter gene, called "ffLuc," a novel fusion protein, for bioluminesence (BLI) imaging in order to track the transplanted cells via MRI from injection site to spinal cord lesion.

"MRI signals from dead cells cannot be distinguished from those of living cells," noted Dr. Nakamura. "To overcome this difficulty, we used a BLI system that tracked only living cells."

After inducing SCI in mouse models, the researchers compared the safety and efficacy results of IL, IT and IV injection methods for transplanting NS/PCs. IL grafting emerged as the best method since the IL-injected animals showed the best survival rates for grafted cells post-injection with no complications in the animals. IT injection showed low animal survival rates. IV injection resulted in no grafted cells migrating to the SCI lesion sites and many of the test animals died of a pulmonary embolism soon after injection.

"Taken together, our findings indicate that the best method for transplanting NS/PCs into the spinal cord injury site is by intralesional injection," summarized Dr. Nakamura.

"It is interesting that this study observed the greatest cell survival after neural stem cell transplantation directly into the lesion site after spinal cord injury" said Dr. Paul Sanberg, coeditor-in-chief of Cell Transplantation and executive director of the University of South Florida Center of Excellence for Aging and Brain Repair. "It will be of considerable importance to determine if this finding translates to increased recovery and also whether this translates to other disorders".

Study Will Test Transplantation Of Gene-Modified Cells To Explore A Potential Cure For HIV Infection


Whether a stem cell transplant using an HIV-infected person's own genetically modified immune cells can become a cure for the disease is the focus of a new $20 million, five-year research grant award announced today by the National Institutes of Health to Fred Hutchinson Cancer Research Center.

Hutchinson Center researchers will use the grant to lead a multifaceted team of scientists and institutions to study whether a person's own stem cells can be engineered to deny HIV entry into the body's blood cells. The researchers also will work to develop tools to eradicate existing reservoirs of infection in the body.

"Funding for research to find a cure for HIV-infected persons represents a paradigm shift," said Keith Jerome, M.D., Ph.D., an expert in viral infections and co-principal investigator of the grant. "HIV has been an incurable, lifelong infection that at best sentences people to a lifetime of complex drug therapies. Now the research field is shifting to address the possibility of a cure. No one would have talked about this approach five years ago."

Jerome is an associate member of the Hutchinson Center's Vaccine and Infectious Disease Division. The other co-principal investigator is stem cell transplant researcher Hans-Peter Kiem, M.D., a member of the Hutchinson Center's Clinical Research Division and recipient of the Center's Jose Carreras/E. Donnall Thomas Endowed Chair for Cancer Research.

"I am particularly excited that we can explore stem cell-based therapies for the cure of HIV," Kiem said. "It is a perfect fit for our institution, which has such a unique history of pioneering stem cell transplantation and research in HIV."

The Hutchinson Center grant was one of three announced today by the National Institute of Allergy and Infectious Diseases, part of the National Institutes of Health, to fund research focused on developing strategies for eradicating HIV infection via its Martin Delaney Collaboratory program.

"I applaud the NIAID's efforts to open up new areas of research funding to find a potential cure for HIV," said Larry Corey, M.D., president and director of the Hutchinson Center. "The work to be done in this grant is groundbreaking and the kind of public/private collaboration that is needed to move the science forward."

The research projects will focus on ways around a major obstacle to long-term control and cure of HIV: the persistence of HIV provirus in reservoirs throughout the bodies of infected persons. The infected cells in these reservoirs are long lived and remain a threat during the lifespan of infected persons. Highly active antiretroviral therapy (HAART), although successful at keeping the spread of HIV under control by inhibiting viral replication, does not eliminate these reservoirs. If a patient discontinues HAART, the virus rebounds.

One approach under investigation is autologous stem cell transplantation, in which the infected patient's own immune cells are genetically modified to be resistant to HIV by eliminating one of the receptors, called CCR5, which HIV needs to infect new cells. This method builds on the Hutchinson Center's long-standing expertise in using transplantation to treat and cure blood cancers and some autoimmune diseases, a Nobel Prize-winning accomplishment that has boosted survival rates from nearly zero to 90 percent for certain types of leukemia.

Stem cell transplantation to eliminate HIV infection has one intriguing precedent.

In 2008, a group of German physicians published results of transplanting an American man who had acute myeloid leukemia and HIV. The so-called "Berlin patient" received a new immune system from donor cells that also carried a rare genetic variation that made them resistant to HIV. The man was able to stop HAART and the virus remained undetectable. However, few stem cell donors have this genetic mutation, so a way must be found to modify the patient's own immune cells to be HIV resistant in order for such a transplant to be more widely available.

A second approach to be studied involves developing DNA-targeting proteins to directly attack the reservoirs of HIV provirus without harming the infected cells themselves. This method would complement the stem cell-based approach and could potentially lead to elimination of the HIV provirus.

In addition to better understanding the biology and virology of gene-modified cells, another goal will be to optimize the combination of stem cell protection and HIV reservoir-purging techniques. Researchers expect to have enough data to begin human clinical trials in five years.

The team of scientists and institutions that will conduct the studies includes Sangamo Biosciences of Richmond, Calif., Beckman Research Institute at City of Hope in Duarte, Calif., the University of Washington and Seattle Children's.

Stem Cell Treatment May Restore Cognitive Function In Patients With Brain Cancer



Stem cell therapy may restore cognition in patients with brain cancer who experience functional learning and memory loss often associated with radiation treatment, according to a laboratory study published in Cancer Research, a journal of the American Association for Cancer Research.

Charles Limoli, Ph.D., a professor in the department of radiation oncology at the University of California, Irvine, said radiation therapy is the standard of care for most brain cancers, but the side effects can be devastating.

"In almost every instance, people experience severe cognitive impairment that is progressive, debilitating and adversely impacts quality of life," he said. "Pediatric cancer patients can experience a drop of up to three IQ points per year."

In the current study, Limoli and colleagues subjected rats to cranial irradiation and followed up two days later with human neural stem cell transplants. A significant proportion of these cells survived and turned into brain cells found at one- and four-month evaluations. Cognitive function significantly improved compared with control rats.

Limoli said the findings of this study were significant, and may help pave the way for a human safety trial to be conducted within a few years if appropriate funding can be secured. Neural stem cells like those used in this study do not present the same ethical questions as embryonic stem cells.
